The dynamic ECM: from fly development to fibrotic disease

My lab has been exploiting the embryonic migration of Drosophila macrophages (hemocytes) as a genetically tractable model to understand the role and regulation of cell migration. However, we recently elucidated the surprising function of hemocytes during fly development: the deposition of extracellular matrix (ECM). I will introduce some new projects in the lab focused on understanding the formation, function, and surprising dynamics of the ECM during growth and morphogenesis of Drosophila tissues.
We have also become interested in understanding the regulation of ECM patterning during scarring. ECM architecture in virtually all types of scarring pathologies becomes highly bundled and aligned over supracellular length scales. We have recently shown that patient fibroblasts isolated from keloid scars maintain a unique capacity to generate an aligned ECM network in 2-D culture and I will discuss the cell and molecular mechanisms driving this long-range ECM anisotropy.
