Signaling mechanisms underlying the robustness and tunability of the plant immune network

The plant immune signaling network is built highly robust against perturbations to internal nodes since it needs to withstand assaults from fast evolving pathogens. It is very difficult to elucidate the mechanisms underlying such a highly robust network using minor perturbations, such as single mutations. Thus we simultaneously or combinatorially perturb major signaling sectors to study the underlying signaling mechanisms, which allows a highly interpretable approach of step-wise network reconstitution. By combining deep and combinatorial perturbations and sector activity measurements, we built a highly predictive, multiple regression model of signal flows in the network. This model showed that during pattern-triggered immunity (PTI), robustness of the network is mainly conferred by inhibition of jasmonate (JA) signaling by ethylene signaling and that tuning of the network response is enabled by variable network input patterns. It also revealed activation of salicylate (SA) signaling by JA signaling, which was experimentally confirmed. Furthermore, a major dichotomy of JA signaling and PAD4 signaling in regulation of immunity was suggested.
