要旨
REM sleep is a distinct brain state associated with vivid dreaming, yet how the brain decides when to enter REM sleep remains unclear. I will present recent work from our lab showing that REM sleep is not simply generated by a local on/off switch, but emerges from slow, coordinated population dynamics across a distributed brainstem network. Using high-density Neuropixels recordings in naturally sleeping mice, we found that brainstem activity evolves along low-dimensional trajectories that gradually prepare the brain for REM sleep and predict when REM will occur. These dynamics also determine whether optogenetic activation of REM-promoting neurons successfully triggers a state transition, suggesting that local circuits act within a broader network state. I will also discuss how this framework extends to the organization of REM sleep itself, including tonic and phasic REM substates. Together, these findings suggest that REM sleep is an emergent network state, prepared during non-REM sleep and dynamically structured.
