The zebrafish (Danio rerio) has figured prominently as a vertebrate model for studying circadian clocks and circadian roles in various life processes. Using several reverse genetic tools including genome-editing techniques TALEN and CRISPR-Cas9, we have generated a library of zebrafish circadian clock mutants. Through characterizing and analyzing these circadian mutants, we have revealed highly conserved circadian functions of these zebrafish clock genes as well as their novel roles in several pathological and physiological processes. My talk will focus on: 1) Per1b’s role in ADHD (Attention Deficit Hyperactivity Disorder), 2) Rev-erbα’s role in autophagy and 3) circadian regulation of hematopoiesis via Ezh2.
